Description
What Is P21 Peptide?
P21 peptide is a modified, synthetic CNTF mimic. In humans, CNTF is a naturally occurring protein that mediates neuron development. The effects of CNTF have mostly been examined in the neurological system, while the peptide has receptors in various parts of the body (for example, bone). It has been demonstrated to stimulate neurotransmitter production and neurite outgrowth. It also protects neurons and the cells that support them from inflammatory assaults. CNTF is known to promote satiety and so reduce food intake, in addition to its neurotrophic benefits.[13][14]
The molecules CNTF and cerebrolysin are not the same. P21 and cerebrolysin are also not the same thing. It is addressed and contrasted with P21 below.[1][2]
Axokine is the brand name for a recombinant form of CNTF. It was studied as a therapy for amyotrophic lateral sclerosis but is no longer available.Interestingly, the body quickly produces antibodies against Axokine, implying that there may be a potential for combining P21 with exogenous CNTF in some conditions, hence increasing CNTF levels while minimizing antibody activity.
How was P21 Peptide Developed?
P21 is a tiny CNTF peptide derivative. Small molecule mimics can mimic some or all of the actions of bigger neutrophic compounds while avoiding the aforementioned adverse effects. P21 was created using a technique known as epitope mapping, which use antibodies to discover target binding sites. Antibodies against CNTF receptor active sites were employed to identify the CNTF binding site in the instance of P21. They were then utilized to determine whether short, synthetic peptides mimicked CNTF binding and, as a result, interfered with antibody binding.
The result was the production of P21, which not only binds to the CNTF receptor, but also crosses the blood-brain barrier and placental/lactational barriers. P21 is a tetra-peptide derived from the most active region of CNTF (amino acid residues 148–151). Admantylated glycine was added to the C-terminal end to increase blood–brain barrier permeability and decrease degradation by exopeptidases. Natural CNTF is too large to cross the blood-brain barrier, has poor plasma stability, an unfavorable pharmacologic profile, and actually promotes the development of anti CNTF-antibodies when administered systemically. Direct administration to the cerebrospinal fluid, while an option, is generally avoided due to pain, risk of infection, and other adverse effects. Unlike full CNTF, P21 has better than 95% stability in artificial gastric juice over 30 minutes, long enough for it to pass through the stomach in most cases. It is roughly 100% stable in the intestine over two hours, which is long enough for it to be absorbed. It is stable in blood plasma for more than 3 hours.
How P21 Works?
P21 peptide stimulates neurogenesis in the dentate gyrus. The dentate gyrus is a component of the hippocampus formation and is thought to be involved in the creation of new episodic memories as well as the exploration and learning that occurs in novel contexts and circumstances. This portion of the brain is also in charge of pre-processing information and increasing pattern separation, which allows animals to distinguish memories.
Neuroscientists are particularly interested in the p21 peptide nootropic for its effects on the gyrus.
According to studies, P21 can have a range of impacts in the body, including elevated BDNF levels, phosphorylated GSK3B levels, and AMPA receptor protein levels.
According to mouse studies, the P21 peptide does not attach to the CNTF receptor, showing that it is not a CNTF homologue. It appears to suppress antibodies and chemicals that neutralize it instead. As a result, the peptide P21 boosts CNTF concentration and mimics its actions.
According to another studies, elevated P21 levels increase BrdU positive cells (synthetic nucleosides) in the dentate gyrus. The same study discovered that P-21 peptide enhances cell proliferation in this location, lending credence to the idea that this proliferation is really increased neurogenesis.[3][4]
What are the benefits of P21 peptide?
Let’s take a look at some of the health benefits of P21, as discovered by researchers. Indeed, it’s an interesting drug which is magnetic to scientists eager to get this approved by the FDA.
Memory
P21 peptide has been shown to improve cognition and protect the central nervous system from internal and external injury. It accomplishes this by promoting the growth of neurons from precursor cells to full neurons. Furthermore, animal studies have demonstrated that P21peptide increases neuronal connectivity, resulting in better learning and memory.
According to the most recent p21 study, this molecule has a lot of advantages, particularly in terms of memory, learning, and general cognitive function. This peptide increases thinking and object discrimination in mice, according to research.
Furthermore, it can increase the levels of GluR1, Synapsin 1, and NR1, all of which are markers of neurogenesis. P21 can enhance these proteins in both sickness and wellness. According to animal studies, p21 is a critical component in learning, memory, and brain function.[5][6]
Alzheimer’s Disease
According to research, P21 therapy enhances activity in the dentate gyrus, which aids in the treatment of Alzheimer’s disease. This is because AD’s normal response to brain injury is to increase activity in the gyrus, but elderly brains lack the ability to do so. Animal studies has revealed that this can be changed with P21 treatment.
According to scientific evidence, P21’s neurotrophic support increases levels of brain-derived neurotrophin-4 and neurotrophic factor. It also reduces the mitogenic effects of growth factor 2. Researchers discovered that administering P21 injections to animal models before to the beginning of AD might function as a preventive measure.
Furthermore, BDNF has been linked to the reduction of enzymes that generate tau and amyloid plaque in Alzheimer’s disease brains. This shows that P21 might be a game-changing Alzheimer’s prevention and therapy.
Cancer
According to new study, p21 regulates all important cellular activities, all of which are involved in the development, growth, and response of malignant cells. That is why scientists are frantically looking into the function of the p21 peptide in cancer treatment.
In fact, several chemotherapeutic drugs have been created to boost p21 activity in order to slow cancer growth.
Due to P21’s inhibitory effect on CDK2 activity, this peptide can prevent cancer cell proliferation – in particular, that of breast cancer. Research studies indicate that the expression of estrogen receptor-a gene is initiated by the activation of p21 peptide hormone.[7][8]
Appetite Suppression
P21 outcomes may, interestingly, include weight decrease. While no direct studies have been conducted to correlate p21 to fat reduction, there is evidence that it may reduce appetite. According to specialists, this is a side consequence of increased CNTF levels boosting alpha-melanocyte stimulating hormone production.
P021 raises levels of alpha-MSH, a molecule linked with lower food consumption, when these levels are raised. So, while it’s not a weight loss medicine in and of itself, there’s reason to suppose it may be one eventually.
P21 side effects and safety
While scientists consider P21 as a safe compound, there are two potential side effects to watch out for, as research on animals has shown, including:
- Tiredness– most neurogenetic compounds product such effects, so this isn’t something to be concerned about.
- Cognitive rebound– this side effect is rare, but it can happen.
Momentarily, there is inadequate research to legalize P21 as an over-the-counter medicine, which is why it is only available to researchers for the sole purpose of research.
P21 appears to have no negative effects in animals with Alzheimer’s disease. That doesn’t mean the nootropic has no adverse effects, but it does imply that if it makes it to clinical trials, the unwanted effects will be minimal. So far, the only adverse effect of P21 peptide seen is decreased anxiety, which is really a beneficial impact.
P21 vs Cerebrolysin
While p21 is frequently referred to as a derivative of cerebrolysin, the two compounds are not the same. p21 peptide is a synthetic counterpart of CNTF, whereas cerebrolysin is a peptide combination in which CNTF is merely one of the components. According to animal research, P21 is made up of only four amino acids from CNTF and is more effective than cerebrolysin.
Cerebrolysin therapy has been abandoned in recent years due to it encouraging the formation of antibodies against CNTF and progressively becoming utterly useless. In fact, it might aggravate the situation.
This product was formerly one of the top alternatives available. It was initially a successful therapy because it was a complicated combination of several brain chemicals. However, it creates large quantities of foreign CNTF, which leads to the creation of autoantibodies, which has negative consequences.
P21 has been shown to capture and reduce autoantibodies, resulting in increased effectiveness of natural CNTF. Studies demonstrate that it does not lose potency with time, which is a very good indicator.[9][10]
All in all, when it comes to the cerebrolysin vs P21 debate, the latter is the clear winner.
